Thousands of people joined the online queue at 09:00 BST in the hope of securing tickets to the festival at Worthy Farm in Somerset, which returns from 23 to 27 June after a fallow year.
Organisers confirmed last month that tickets for the weekend were at their highest price ever - £408, including a £5 booking fee per ticket - an increase of £29.50 from the last event in 2025.
Headliners have yet to be announced but organiser Emily Eavis previously said the team was "very excited" and things were "coming together really well".
Glastonbury posted on Instagram shortly after 09:40 to announce that tickets had sold out, and notified those who were in the queue.
"Thank you to everyone who bought one and we're sorry to those of you who missed out on a morning where demand greatly exceeded supply," the post said.
"There will be a resale of any cancelled or returned tickets in Spring 2027."

The first round of tickets, which were for coach packages, went up for sale on Thursday and sold out in less than half an hour.
Despite the increase in cost, Zoe Scott, who has been to the festival twice, said "the price is totally worth it".
"You're getting to see thousands of acts, there's over 100 stages," she said.
"It's such a vibe at Glastonbury- everyone's so happy and free-spirited. You're just on a constant high from friends, music and the sun is usually shining".
